Teach PCR, gel electrophoresis, and bioinformatics with this DNA barcoding lab activity in which students determine the species of a fish based on the DNA sequence of its cytochrome c oxidase I (COI) gene. Students first extract DNA from tissue, use PCR to amplify the DNA, and perform agarose gel electrophoresis to verify PCR products. Next, after receiving DNA sequencing results from a third party (sequencing services not included), students then use bioinformatics to genetically identify the species using the sequence obtained. The unique COI sequence produced for a given sample is considered its DNA barcode.
